MOD-801 is a long-acting analog of glucagon-like peptide-1 (GLP-1) developed by OPKO Biologics (formerly PROLOR Biotech) for the treatment of type 2 diabetes. The drug utilizes proprietary Carboxy-Terminal Peptide (CTP) technology, which involves fusing the C-terminal peptide of the human chorionic gonadotropin (hCG) beta subunit to the GLP-1 molecule. This modification is designed to significantly extend the half-life of the peptide by reducing renal clearance and protecting it from metabolic degradation, potentially allowing for once-weekly or less frequent administration. As a GLP-1 receptor agonist, MOD-801 stimulates insulin secretion in a glucose-dependent manner, suppresses glucagon secretion, and slows gastric emptying to improve glycemic control. Development of MOD-801 was discontinued in the early stages of the pipeline.
