Overview

MOD-E is a small-molecule inhibitor of diacylglycerol kinase alpha (DGKα) currently under development by Alivexis as a cancer immunotherapy. DGKα is an enzyme that converts diacylglycerol (DAG) into phosphatidic acid (PA); since DAG is a key secondary messenger for T-cell receptor signaling, its depletion by DGKα leads to T-cell anergy. By inhibiting DGKα, MOD-E aims to restore and enhance T-cell activation and effector function within the immunosuppressive tumor microenvironment. Preclinical data suggests that MOD-E may act synergistically with anti-PD-1 antibodies to improve anti-tumor efficacy. The program is currently in the final preclinical stage, focused on clinical candidate identification.
