MOD016

MOD016 is a preclinical small-molecule oncology drug candidate being developed by Modifi Bio as part of a platform of DNA-targeting agents designed to induce cytotoxic DNA lesions in tumor cells with a potentially improved therapeutic index over existing alkylating chemotherapies. Limited public information from investor materials indicates that MOD016 is being profiled extensively in vivo, including dose, regimen, and therapeutic index optimization, with the goal of advancing to an IND filing; its activity has been explored in intracranial MGMT-deficient and mismatch repair–deficient glioma patient-derived xenograft models where related molecules from the same platform have shown robust survival benefits. The precise molecular target, chemical scaffold, and mechanism of action of MOD016 have not yet been disclosed, but it appears to represent a next-generation, small-molecule, DNA-modifying antineoplastic agent intended for the treatment of solid tumors.
