MODI-1 + pembrolizumab is an investigational combination regimen consisting of MODI-1, a peptide-based cancer vaccine, and pembrolizumab, a monoclonal antibody checkpoint inhibitor targeting PD-1. MODI-1 is composed of citrullinated and homocitrullinated peptides designed to stimulate an anti-tumor immune response. Pembrolizumab blocks the PD-1 receptor, enhancing T-cell mediated immune responses against tumors. The combination aims to leverage MODI-1's vaccine-driven T-cell activation alongside checkpoint inhibition by pembrolizumab, potentially improving immune recognition and destruction of cancer cells. The regimen is currently being investigated primarily for solid tumors including head and neck cancer, renal cancer, and triple negative breast cancer[5].
