MODI-2 is a therapeutic cancer vaccine developed by Scancell using its proprietary Moditope® platform. It is designed to target a unique class of post-translational modifications (PTMs) known as homocitrullination, which involves the conversion of lysine residues to homocitrulline. This process is upregulated in the stressed environment of highly immunosuppressed tumors. MODI-2 utilizes the SNAPvax™ technology to co-deliver homocitrullinated peptide antigens alongside TLR-7/8a adjuvants within self-assembling nanoparticles. These nanoparticles are engineered to prime tumor-killing CD4 T cells. The vaccine targets homocitrullinated epitopes derived from several proteins, including vimentin, aldolase, cytokeratin 8, immunoglobulin binding protein (BiP), nucleophosmin (NPM), α-enolase, and heat shock protein (HSP-60). Preclinical studies have demonstrated strong anti-tumor immunity in models of melanoma, colorectal cancer, and breast cancer.
