Overview

Modified annexin A5 refers to a series of engineered recombinant protein variants developed by Rubicon Biotechnology, licensed from MosaMedix B.V., designed for tumor-targeted immunotherapy. Annexin A5 is a human protein that naturally binds with high affinity to phosphatidylserine (PS), a phospholipid typically sequestered inside healthy cells but externalized on the surface of apoptotic cells and tumor-associated cells (including tumor cells and vascular endothelial cells). In the tumor microenvironment, externalized PS acts as a potent immunosuppressive signal. Rubicon's modified variants are specifically engineered to be non-internalizing, ensuring they remain on the cell surface to mask PS and potentially reverse its immunosuppressive effects, thereby stimulating an anti-tumor immune response. The program is currently in the preclinical stage of development.
