Overview

Modified B16-BL6 vaccine is a research-stage cancer vaccine based on B16-BL6 murine melanoma cells that have been physically (hydrostatic pressure) and chemically (cell-surface protein crosslinking) altered to enhance their immunogenicity. These modifications increase presentation of tumor-associated antigens and major histocompatibility complex (MHC) molecules on the cell surface, making the vaccine more effective at inducing cell-mediated immunity in vivo. Immunization with modified cells generates a specific systemic immune response against B16-BL6 melanoma, stimulates splenocyte proliferation, and increases susceptibility of the tumor cells to cytotoxic lymphocytes while remaining resistant to natural killer cells. Development and study are preclinical, primarily in mouse models for melanoma immunotherapy. The vaccine does not contain additional adjuvants such as interleukin-2, as addition of IL-2 did not increase efficacy in studies. The approach demonstrates potential for developing effective tumor vaccines via physical and chemical antigenic modulation[1].
