A genetically engineered variant of Interleukin-37 (IL-37), an anti-inflammatory cytokine of the IL-1 family. IL-37 naturally suppresses innate and adaptive immune responses by binding to the IL-18 receptor alpha (IL-18Rα) and the orphan receptor IL-1R8 (SIGIRR). This specific modified variant was designed to overcome the limitations of recombinant wild-type IL-37, such as rapid degradation and suboptimal receptor interaction, by enhancing its stability and receptor affinity. In preclinical murine models of allergic asthma (house dust mite and ovalbumin models), intranasal administration of the modified IL-37 variant significantly reduced airway inflammation, eosinophilia, and mucus production, demonstrating superior efficacy compared to the natural cytokine. It is being investigated as a potential biologic therapy for allergic asthma.
