Overview

Modified p53 neoantigen peptides are therapeutic cancer vaccine candidates designed to target mutations in the p53 tumor suppressor gene, which is frequently altered in human malignancies. Developed by XtalPi (QuantumPharm) using a proprietary AI-driven MHC-I presentation prediction model and computer-assisted saturation mutagenesis, these peptides are engineered with specific single amino acid substitutions. This 'heteroclitic' design approach is intended to increase the binding affinity and presentation probability of the peptides by specific HLA alleles, including HLA-A*02:01, HLA-A*03:01, and HLA-A*11:01. By enhancing the presentation score, these modified peptides aim to overcome the typically low immunogenicity of native p53 neoantigens and induce a robust CD8+ T-cell response that cross-reacts with the original mutant p53 proteins expressed on tumor cells. Preclinical studies in humanized mouse models have demonstrated that these AI-designed peptides can effectively reactivate cellular immunity against native p53 neoantigens.
