Overview

The modified vaccinia Ankara (MVA) HIV vaccine (specifically the MVA-CMDR construct) is a recombinant viral vector-based vaccine developed by the U.S. Military HIV Research Program (MHRP), the Walter Reed Army Institute of Research (WRAIR), and the National Institute of Allergy and Infectious Diseases (NIAID). It has been clinically evaluated by the Swedish Institute for Infectious Disease Control (SMI) and the Karolinska Institutet. The vaccine utilizes a highly attenuated, replication-deficient Modified Vaccinia Ankara virus as a vector to deliver HIV-1 antigens, specifically expressing subtype E Env (gp150) and subtype A Gag/Pol from Thai isolates CM235 and CM240. In the HIVIS 01/02 and HIVIS 03 trials, the vaccine was evaluated as a heterologous booster following priming with a multigene HIV-1 plasmid DNA vaccine (HIVIS-DNA) in healthy, HIV-negative adults to elicit both robust humoral and cellular immune responses for the prevention of HIV infection.
