modotuximab

Modotuximab is a chimeric monoclonal antibody that targets the epidermal growth factor receptor (EGFR). It is one of the two component antibodies of Sym004, a mixture developed for enhanced anti-EGFR activity. Modotuximab has been investigated as monotherapy and in combination with futuximab or chemotherapy agents for the treatment of metastatic colorectal cancer, particularly in patients with KRAS/NRAS and BRAF wild-type tumors who have acquired resistance to prior anti-EGFR monoclonal antibody therapy. The mechanism involves binding to distinct non-overlapping epitopes on EGFR, leading to receptor internalization and degradation, thereby inhibiting downstream signaling pathways involved in tumor cell proliferation and survival[8][6]. Clinical trials have evaluated its safety and efficacy both alone and as part of combination regimens.
