ModraDoc 005

ModraDoc 005 is an investigational oral tablet formulation of the taxane chemotherapy agent docetaxel. Developed by researchers at the Antoni van Leeuwenhoek Ziekenhuis (Netherlands Cancer Institute), it is part of the ModraDoc series (ModraDoc001–006) designed to overcome the poor oral bioavailability of docetaxel. To achieve therapeutic systemic exposure, ModraDoc 005 is co-administered with ritonavir, which acts as a booster by inhibiting the CYP3A4 metabolic enzyme and the P-glycoprotein efflux transporter in the gastrointestinal tract. The drug's mechanism of action involves binding to and stabilizing microtubules, which prevents their disassembly, leading to cell cycle arrest and apoptosis in rapidly dividing cancer cells. It has been evaluated in Phase I clinical trials for the treatment of various malignant solid tumors.
