ModraPac001 (MP1) is an investigational oral solid dispersion formulation of the taxane chemotherapy agent paclitaxel, developed by the Netherlands Cancer Institute - Antoni van Leeuwenhoek Hospital. The formulation consists of paclitaxel, polyvinylpyrrolidone (PVP) K30, and sodium lauryl sulfate (SLS) in a 1:9:1 weight ratio, designed to improve the solubility and bioavailability of paclitaxel when administered orally. It is typically co-administered with ritonavir, a CYP3A4 inhibitor, which acts as a pharmacokinetic booster to further enhance oral absorption. ModraPac001 is specifically designed for daily low-dose metronomic (LDM) chemotherapy, aiming to provide continuous exposure to the drug while minimizing toxicity. Its primary mechanism of action is microtubule stabilization, leading to cell cycle arrest and apoptosis in cancer cells. It is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors.
