Overview

modRNA-IL12 is a modified messenger RNA (mRNA) therapeutic designed to encode Interleukin-12 (IL-12), a potent proinflammatory cytokine. The drug is typically encapsulated in lipid nanoparticles (LNPs), often utilizing specific cationic lipids such as TT3, which facilitate intracellular delivery and are capable of triggering immunogenic cell death (ICD). Upon delivery, primarily through intratumoral injection, the modRNA is translated into IL-12 protein within the tumor microenvironment. This localized production of IL-12 activates natural killer (NK) cells and cytotoxic T lymphocytes, promoting a robust anti-tumor immune response while avoiding the systemic toxicities associated with recombinant IL-12 protein therapy. Preclinical studies have demonstrated its efficacy in reducing tumor burden and improving survival in difficult-to-treat melanoma models.
