Overview

Modzatinib (ATI-2138) is an orally bioavailable small molecule kinase inhibitor developed by Aclaris Therapeutics. It is designed to covalently inhibit Janus kinase 3 (JAK3) and interleukin-2-inducible T-cell kinase (ITK). By targeting these two kinases, modzatinib aims to modulate both the JAK-STAT signaling pathway and T-cell receptor signaling, which are central to the pathogenesis of various inflammatory and autoimmune diseases. It is currently being evaluated in Phase 2 clinical trials for the treatment of moderate-to-severe atopic dermatitis and various forms of lichen planus, including oral, cutaneous, and lichen planopilaris.
