Mofegiline (MDL 72974A) is a **potent, selective, irreversible inhibitor of monoamine oxidase B (MAO-B)**, also characterized as a mechanism-based (enzyme-activated) inhibitor. It also inhibits semicarbazide-sensitive amine oxidases (SSAO/VAP-1) with high potency. Mofegiline acts by covalently binding to the active site of MAO-B, leading to long-lasting inhibition. It was developed for neuroprotective potential in **Parkinson’s disease** and possibly other neurodegenerative disorders, and shows oral bioavailability and long-lasting inhibition of platelet MAO-B. Clinical studies showed activity in Parkinson’s patients, but it was never widely marketed. Mofegiline also has anti-inflammatory effects observed in vitro and in vivo[1][2][3][4][5].
