Drug intelligence / Profile preview
MOG mRNA Tol-cLNP vaccine is an investigational tolerizing mRNA-lipid nanoparticle (LNP) vaccine developed by etherna in collaboration with Hasselt University for the treatment of multiple sclerosis (MS). The vaccine consists of mRNA encoding myelin oligodendrocyte glycoprotein (MOG) (specifically MOG27-63) encapsulated in customized tolerizing lipid nanoparticles (Tol-cLNP). It is designed to target antigen-presenting cells (APCs) in the liver and spleen to present the MOG antigen in an immune-silent manner. This induces antigen-specific tolerance by promoting a homeostatic APC phenotype and driving the proliferation of MOG-specific T cells into an exhausted, anti-inflammatory phenotype with increased IL-10 expression, thereby suppressing autoimmune responses without causing generalized immunosuppression.
