Overview

MOG35-55 peptide is a synthetic peptide representing the immunodominant epitope of myelin oligodendrocyte glycoprotein (MOG), spanning amino acid residues 35 to 55 (sequence: Met-Glu-Val-Gly-Trp-Tyr-Arg-Ser-Pro-Phe-Ser-Arg-Val-Val-His-Leu-Tyr-Arg-Asn-Gly-Lys). It is primarily utilized as a research tool to induce experimental autoimmune encephalomyelitis (EAE) in mouse models, which serves as a standard preclinical model for multiple sclerosis (MS) and other autoimmune demyelinating diseases. Mechanistically, the peptide acts as an agonist for myelin-specific CD4+ T cells by binding to the T cell receptor (TCR) when presented by MHC class II molecules (specifically I-Ab in C57BL/6 mice). Beyond its use in disease induction, MOG35-55 has been investigated in various modified or conjugated forms, such as mannan-conjugated or nanoparticle-encapsulated versions, as a potential tolerogenic immunotherapy designed to induce antigen-specific immune tolerance and suppress the autoimmune response in MS.
