Overview

Mogamulizumab + nivolumab is a combination of two monoclonal antibodies used in clinical trials for the treatment of locally advanced or metastatic solid tumors. Mogamulizumab is a humanized monoclonal antibody targeting CC chemokine receptor 4 (CCR4), which is expressed on certain T cells, including regulatory T cells (Tregs). By binding to CCR4, mogamulizumab inhibits chemokine-mediated signaling and induces antibody-dependent cell-mediated cytotoxicity against CCR4-positive cells. Nivolumab is a fully human IgG4 monoclonal antibody that targets programmed cell death protein 1 (PD-1) on T-cells, blocking its interaction with PD-L1 and PD-L2 ligands. This blockade restores tumor-specific T-cell responses by preventing tumor-induced immune suppression. The combination was evaluated in phase I/II studies for safety and antitumor activity but did not demonstrate enhanced efficacy compared to monotherapy; however, it showed acceptable tolerability[1][2][5].
