Overview

moIL-2 is a pegylated, mutated mouse interleukin-2 (IL-2) mutein developed by Synthekine. It is designed as part of an orthogonal receptor/ligand system to selectively activate engineered T cells that express a corresponding mutated IL-2 receptor beta (moRβ). By specifically targeting the orthogonal receptor and exhibiting minimal activity toward the wild-type IL-2 receptor, moIL-2 aims to provide the potent stimulatory effects of IL-2—such as enhanced T-cell proliferation and persistence—while avoiding the systemic toxicities and off-target immune activation (e.g., NK cells and regulatory T cells) typically associated with high-dose IL-2 therapy. In preclinical models, moIL-2 has demonstrated the ability to support adoptive T-cell therapy (ACT) efficacy in lymphoreplete environments, potentially eliminating the need for toxic lymphodepletion regimens.
