Overview

Molecule 1102-39 is a bone-targeted prodrug of methotrexate developed by Atlanthera for the treatment of bone-related inflammatory and oncologic conditions. It utilizes a hydroxybisphosphonate (HBP) vector to selectively deliver the antifolate therapy to bone and joint tissues by binding to the bone mineral matrix. This targeted delivery mechanism aims to achieve high local concentrations of the drug while reducing systemic exposure and associated toxicities. Once localized, the methotrexate moiety is released to inhibit dihydrofolate reductase (DHFR), thereby disrupting folate metabolism and exerting anti-inflammatory or anti-proliferative effects. As of late 2024, the molecule is in the preclinical stage of development, with published data from in vitro and in vivo rodent models.
