MOMA-341 is an oral, potent, and selective covalent inhibitor of Werner helicase (WRN) with a novel chemical scaffold. Developed by MOMA Therapeutics using their proprietary KNOMATIC platform, it is designed to target cancers characterized by microsatellite instability-high (MSI-H) or DNA mismatch repair deficiency (dMMR), including colorectal, gastric, and endometrial cancers. The drug is being evaluated as both monotherapy and in combination with chemotherapy (such as irinotecan) or immunotherapy in advanced or metastatic solid tumors. Its mechanism involves inhibiting the WRN protein—a key player in DNA repair—making it particularly relevant for tumors dependent on this pathway due to underlying genetic instability. As of mid-2025, MOMA-341 has entered Phase 1 clinical trials to assess safety, tolerability, pharmacokinetics/pharmacodynamics, and preliminary efficacy[1][2][3][4][5][6][7][8].
