Overview

Monalizumab is a humanized IgG4 monoclonal antibody that acts as an immune checkpoint inhibitor targeting the inhibitory receptor NKG2A (also known as CD94/NKG2A or KLRC1) on cytotoxic CD8+ T cells and natural killer (NK) cells. By blocking the interaction between NKG2A and its ligand HLA-E, which is often overexpressed on tumor cells, monalizumab prevents tumor-mediated inhibition of these immune effector cells. This blockade enhances both innate and adaptive anti-tumor responses by restoring the cytotoxic activity of NK and CD8+ T cells against cancer. Monalizumab is being developed primarily for oncology indications, including non-small cell lung cancer (NSCLC), small cell lung cancer (SCLC), head and neck squamous cell carcinoma, chronic lymphocytic leukemia, gynecologic cancers, colorectal cancer, esophageal/gastroesophageal junction cancers, among others[1][3][4][5].
