mONCR-171 is a preclinical **mouse surrogate** of ONCR-177, an engineered **oncolytic herpes simplex virus type 1** immunotherapy developed by **ONCR** for cancer. It is a multi-transgene HSV-1 vector designed to selectively replicate in and lyse tumor cells while stimulating systemic antitumor immunity. The construct is armed with five immunomodulatory payloads, including **interleukin-12**, **C-C motif chemokine ligand 4**, **Fms-related tyrosine kinase 3 ligand**, and antagonistic biologics directed against **programmed cell death protein 1** and **cytotoxic T-lymphocyte associated protein 4**. It also incorporates tumor-selectivity and safety features including **tissue-specific microRNA attenuation** and **UL37 mutation** to reduce replication in normal cells while preserving oncolytic activity in tumor tissue. In preclinical bilateral syngeneic mouse tumor models such as A20 lymphoma, B16F10 melanoma, and MC38 colon carcinoma, mONCR-171 showed both local and abscopal antitumor effects associated with increased infiltration of activated NK cells, CD8 T cells, CD4 T cells, and dendritic cells.
