Overview

Monocyte-derived dendritic cell (MoDC) vaccines are a form of autologous cellular immunotherapy used primarily in oncology. The process involves harvesting monocytes from a patient's peripheral blood via leukapheresis, followed by ex vivo differentiation into dendritic cells using cytokines such as GM-CSF and IL-4. These immature DCs are then loaded with tumor-associated antigens—which can be in the form of synthetic peptides, tumor lysates, or nucleic acids—and matured using inflammatory stimuli, such as Toll-like receptor (TLR) agonists. Once re-infused into the patient, these professional antigen-presenting cells migrate to lymphoid organs to prime and activate tumor-specific CD8+ cytotoxic T cells and CD4+ helper T cells, thereby orchestrating a systemic immune response against the malignancy.
