Monoiodoacetate is a small molecule inhibitor of glycolysis, specifically targeting the enzyme glyceraldehyde-3-phosphate dehydrogenase, resulting in disruption of cellular glycolysis and subsequent cell death, especially in chondrocytes. It is most commonly utilized in preclinical research to induce osteoarthritis-like cartilage degeneration and pain in animal models via intra-articular injection. The compound's action mimics features of human osteoarthritis, including cartilage degeneration and bone alterations, by causing chondrocyte cell death and triggering an inflammatory cascade in the injected joint. Because of its mechanism, it is highly toxic if administered systemically[1][3][5][7].
