Monomeric anti-FRα IgG1 is a human monoclonal antibody targeting folate receptor alpha (FRα), isolated from a human phage display library. Developed by Medicovestor and Sanyou Biopharmaceuticals, this antibody serves as the structural building block for the ADoBind platform. In this platform, the monomeric IgG1 is engineered with an S444C mutation in the heavy chain to allow disulfide-bond crosslinking into a tetravalent format. As a standalone monomeric entity, the antibody binds to FRα-expressing cells and mediates antibody-dependent cellular cytotoxicity (ADCC). In preclinical studies, it has been used as a comparative control for the tetravalent antibody and the antibody-drug conjugate (ADC) MC001, demonstrating lower avidity, lower serum concentrations (AUC), and reduced anti-tumor potency compared to the multivalent versions in models of pancreatic and ovarian cancer.
