A monospecific monoclonal antibody targeting the integrin α5β1 receptor, also known as the fibronectin receptor. Integrin α5β1 is a heterodimeric cell surface protein that plays a critical role in cell-extracellular matrix interactions, mediating processes such as cell adhesion, migration, and survival. In oncology, α5β1 is frequently upregulated in tumor cells and the tumor vasculature, contributing to angiogenesis, metastasis, and treatment resistance. In the context of prostate cancer research, specifically basal-type prostate cancer (BTPC), monospecific α5β1 antibodies are utilized to investigate the role of this integrin in chemotaxis and clonogenic survival. While these antibodies can inhibit α5β1-mediated signaling, research presented at AACR 2025 suggests that bispecific antibodies targeting both α5β1 and αv integrins may provide superior therapeutic outcomes by inducing lysosomal degradation of the targets and promoting immunological control of the tumor through the recruitment of natural killer (NK) cells and macrophages.
