A monospecific MUC1 antibody-drug conjugate (ADC) is a targeted cancer therapeutic designed to deliver a cytotoxic payload directly to cells expressing Mucin-1 (MUC1). MUC1 is a transmembrane glycoprotein that is frequently overexpressed and exhibits aberrant glycosylation in various epithelial malignancies, including breast, lung, and pancreatic cancers. The ADC consists of a monoclonal antibody specific to the MUC1 antigen covalently linked to a potent cell-killing agent. While MUC1 is a well-validated tumor-associated antigen, the effectiveness of monospecific targeting can be limited by the heterogeneity of MUC1 expression and the shedding of the MUC1 extracellular domain into the systemic circulation, which can act as a decoy for the therapeutic antibody. In the context of drug development, monospecific MUC1 ADCs are often utilized as benchmarks or controls to demonstrate the superior binding, internalization, and anti-tumor activity of next-generation multi-specific agents, such as bispecific EGFRxMUC1 ADCs.
