The monovalent norovirus vaccine refers to oral tablet vaccine candidates developed by Vaxart, specifically VXA-G1.1-NN (targeting the GI.1 Norwalk strain) and VXA-G2.4-NS (targeting the GII.4 strain). These vaccines utilize a non-replicating adenovirus serotype 5 (Ad5) vector platform that is E1/E3-deleted. The vector is engineered to express the full-length VP1 capsid protein of the norovirus, which serves as the primary antigen. The vaccine is formulated into an enteric-coated tablet designed to bypass the stomach and release in the small intestine, where it targets gut-associated lymphoid tissue (GALT). It incorporates a double-stranded RNA (dsRNA) adjuvant to stimulate a robust mucosal and systemic immune response. This platform is notable for being room-temperature stable and providing a needle-free alternative to traditional vaccines.
