MOPEVAC-Las (also known as MOPEVAC-LAS or MOPEVACLASV) is a recombinant, live-attenuated viral vector vaccine candidate developed by the Institut Pasteur for the prevention of Lassa fever. The vaccine is built on the MOPEVAC (Mopeia vaccine) platform, which utilizes a genetically modified, hyperattenuated strain of the non-pathogenic Mopeia virus (MOPV). To construct MOPEVAC-Las, the MOPV envelope glycoprotein (GPC) gene is replaced with the GPC gene of the Lassa virus (LASV). Additionally, the vector is hyperattenuated by introducing six mutations into the exoribonuclease (ExoN) domain of the MOPV nucleoprotein (NP), which prevents the virus from suppressing the host's type I interferon (IFN) response. This modification enhances both safety and immunogenicity, promoting a robust innate and adaptive immune response (including neutralizing antibodies and T-cell activation) after a single dose. MOPEVAC-Las is currently being evaluated in a Phase 1 clinical trial in healthy volunteers.
