Drug intelligence / Profile preview
MOPEVAC-Las is a live-attenuated recombinant vaccine candidate developed by the Institut Pasteur for the prevention of Lassa fever. The vaccine is built on the proprietary MOPEVAC (Mopeia vaccine) platform, which utilizes a genetically engineered, hyperattenuated Mopeia virus (MOPV) backbone. MOPV is naturally non-pathogenic to humans, and the MOPEVAC platform is further hyperattenuated by introducing six mutations in the exoribonuclease (ExoN) domain of the nucleoprotein (NP) to prevent the virus from suppressing the host's type I interferon response. To target Lassa virus (LASV), the glycoprotein complex (GPC) gene of MOPV is replaced with the GPC gene of LASV (specifically from the Josiah strain). MOPEVAC-Las is designed to elicit both robust humoral (neutralizing antibodies) and cell-mediated (CD4+ and CD8+ T-cell) immune responses. It is currently being evaluated in a Phase 1 clinical trial (FIRST-IN-MOP) in healthy volunteers.
