MORAb-022 is a fully human IgG1 monoclonal antibody designed to target and neutralize granulocyte-macrophage colony-stimulating factor (GM-CSF). Developed by Morphotek (a subsidiary of Eisai) in collaboration with the Ludwig Institute for Cancer Research, the antibody was engineered using Morphotek's proprietary Human Morphotek (HuMab) technology. GM-CSF is a pro-inflammatory cytokine that plays a critical role in the differentiation, activation, and survival of myeloid cells, such as macrophages and neutrophils, which are key drivers of tissue damage in autoimmune and inflammatory diseases. By binding to GM-CSF, MORAb-022 prevents the cytokine from interacting with its cognate receptor, thereby inhibiting the downstream signaling pathways that lead to chronic inflammation. While primarily investigated for the treatment of rheumatoid arthritis and other inflammatory conditions, it was also considered for potential applications in oncology to modulate the tumor microenvironment. Clinical development reached Phase 1, but the candidate is no longer listed in active development pipelines.
