Overview

A MORF4L1 inhibitor is a therapeutic agent designed to block the activity of Mortality Factor 4-Like 1 (MORF4L1), also known as MRG15. MORF4L1 is a highly conserved protein that serves as a critical adapter component in both histone acetyltransferase (HAT) and histone deacetylase (HDAC) complexes, thereby regulating chromatin remodeling, DNA damage repair, and gene transcription. The small molecule thrombin inhibitor argatroban has been identified as a potent MORF4L1 antagonist, binding to the MRG domain and disrupting its interaction with partner proteins. Research indicates that inhibiting MORF4L1 can sensitize hepatocellular carcinoma (HCC) cells to radiotherapy by activating the cGAS-STING signaling pathway and can suppress the growth of T-cell acute lymphoblastic leukemia (T-ALL) by downregulating cholesterol biosynthesis genes such as HMGCR and HMGCS1. Additionally, MORF4L1 inhibition has been studied for its potential to reduce lung epithelial cell death following Pseudomonas aeruginosa infection.
