Overview

Morfisudine (HEC50693, also referred to as morvisudine) is an orally bioavailable small molecule that acts as a capsid assembly modulator (CAM) for the treatment of chronic hepatitis B virus (HBV) infection. Developed by HEC Pharm (Dongguan Chang'an Dongyangguang New Drug Research and Development), it belongs to the core protein allosteric modulator (CpAM) class. Morfisudine specifically targets the HBV core protein (HBcAg), binding to an allosteric site to disrupt the assembly of viral nucleocapsids. This interference leads to the formation of empty or aberrant capsids and prevents the encapsidation of pre-genomic RNA (pgRNA), thereby inhibiting viral DNA synthesis and replication. Morfisudine has been evaluated in Phase 1 and Phase 2 clinical trials, demonstrating the potential to reduce HBV DNA and RNA levels in patients, often studied in combination with standard-of-care nucleoside/nucleotide analogues.
