Morgiduxostat (TP-0462) is an orally bioavailable, small molecule inhibitor of hematopoietic progenitor kinase 1 (HPK1), also known as MAP4K1. HPK1 is a negative regulator of T-cell, B-cell, and dendritic cell activation, and its inhibition is intended to enhance anti-tumor immunity by promoting T-cell receptor signaling and overcoming immunosuppression within the tumor microenvironment. Developed by Taiho Pharmaceutical, morgiduxostat is currently being investigated in clinical trials for the treatment of advanced solid tumors, both as a monotherapy and in combination with other immunotherapies such as checkpoint inhibitors. By targeting a key intracellular checkpoint, it represents a novel approach to cancer immunotherapy designed to augment the body's natural immune response against malignant cells.
