Morinamide is a small molecule, second-line antituberculosis agent classified as a pyrazinecarboxamide. It is used in the treatment of tuberculosis and acts as a prodrug for pyrazinamide. Its primary mechanism of action involves inhibition of mycolic acid synthesis by targeting fatty acid synthase II (FAS-II), which disrupts the integrity of the Mycobacterium tuberculosis cell wall, leading to bacteriostatic and bactericidal effects. The drug’s efficacy is pH-dependent, with optimal activity at acidic pH (around 5.6). Morinamide is typically administered orally as its hydrochloride salt.
