This is a combination of three extended-release opioid analgesics: morphine sulfate, oxycodone, and oxymorphone. Each drug acts primarily as an agonist at the mu-opioid receptor, producing analgesia by inhibiting pain signaling in the central nervous system. Morphine and its derivatives are considered prototypical opioids; oxycodone also has activity at kappa and delta opioid receptors, while oxymorphone is a potent mu-opioid receptor agonist with higher lipophilicity than morphine, allowing for more efficient blood-brain barrier penetration[2][5][6]. All three drugs are indicated for the management of severe pain requiring continuous, long-term opioid treatment when alternative options are inadequate[2][3][4]. These agents have similar adverse effect profiles typical of opioids (e.g., constipation, nausea/vomiting, sedation)[2]. Extended-release formulations provide sustained plasma concentrations to allow 12-hour or longer dosing intervals.
