MOS102 is a small molecule MDM2 inhibitor being developed by Mosaic Therapeutics for the treatment of p53 wild-type (WT) myelofibrosis. By inhibiting the E3 ubiquitin-protein ligase MDM2, MOS102 prevents the degradation of the p53 tumor suppressor protein, thereby restoring p53-mediated cell cycle arrest and apoptosis in malignant cells. The drug is currently in IND-enabling development, with a Phase 1b clinical trial planned for the first half of 2027. It is designed for use as a combination therapy to enhance therapeutic efficacy in myelofibrosis patients who retain wild-type p53 function.
