MOS1SIP + MOS2SIP + M3SIP8 + MPLA-5 is an experimental HIV-1 vaccine candidate developed by the International AIDS Vaccine Initiative (IAVI). The vaccine is a trivalent formulation containing three computationally designed mosaic HIV-1 envelope (Env) trimer immunogens—MOS1SIP, MOS2SIP, and M3SIP8. These mosaic proteins are engineered to represent a broad range of global HIV-1 genetic diversity, aiming to elicit cross-reactive neutralizing antibodies. The immunogens are stabilized in their native-like trimeric state to better present key epitopes to the immune system. The formulation includes MPLA-5, a liposomal adjuvant containing Monophosphoryl Lipid A, which serves as a Toll-like receptor 4 (TLR4) agonist to stimulate innate immune pathways and enhance the adaptive response. Clinical development reached Phase 1, where it was intended to study bolus versus fractionated dosing in people living with HIV (PLWH), but the trial was withdrawn due to funding cuts.
