Mos3.1 is a rationally designed, prefusion-stabilized, native-like HIV-1 envelope (Env) glycoprotein gp140 trimer. It was developed as part of the European AIDS Vaccine Initiative (EAVI2020) to address the significant global diversity of HIV-1. Unlike traditional immunogens based on a single viral isolate, Mos3.1 is a "mosaic" protein whose sequence is computationally optimized to include segments from various viral strains, thereby maximizing the coverage of potential T-cell and B-cell epitopes. In clinical experimental medicine studies, Mos3.1 is typically administered as a booster injection following a "consensus" Env prime (such as ConM or ConS) to focus the immune response toward conserved neutralizing epitopes and broaden the breadth of the elicited antibody repertoire.
