Mosaic gp140 is a recombinant protein immunogen designed as part of investigational HIV vaccine regimens. It is an engineered version of the HIV-1 envelope glycoprotein 140 (gp140), constructed using "mosaic" sequences that combine elements from multiple global HIV subtypes to broaden immune coverage. Mosaic gp140 has been used in combination with other components such as clade C gp140 and adenovirus serotype 26 vectors (Ad26.Mos4.HIV) in clinical trials for preventive HIV vaccines. The protein is typically administered with an adjuvant (aluminum phosphate) to enhance immunogenicity. Preclinical and early clinical studies showed that mosaic gp140 can elicit high binding antibody titers and cross-clade neutralizing antibodies, though large efficacy trials did not demonstrate sufficient protection against HIV infection[1][2][3][5]. Mosaic-based vaccine regimens were developed by Janssen Vaccines & Prevention B.V., often in collaboration with partners including the National Institutes of Health, Beth Israel Deaconess Medical Center, International AIDS Vaccine Initiative, U.S. Military HIV Research Program, and Johnson & Johnson[1][3][4]. Development for prevention of HIV infection has been discontinued after Phase 3 trials failed to meet efficacy endpoints[1][4].
