Overview

Mosaic HIV-1 envelope trimer immunogens are a series of engineered recombinant proteins designed to address the high genetic diversity of HIV-1. These immunogens utilize "mosaic" sequences—computationally optimized sequences designed to maximize the representation of potential T-cell and B-cell epitopes from diverse global HIV-1 strains. They are typically presented in the **SOSIP** format, which stabilizes the envelope (Env) protein in its native-like, prefusion trimeric conformation through a disulfide bond (SOS) and a proline mutation (IP). By mimicking the structure of the functional HIV-1 spike, these trimers aim to elicit broadly neutralizing antibodies (bNAbs) against conserved sites of vulnerability, such as the CD4 binding site, the V3-glycan patch, and the apex. These immunogens are often administered as a cocktail (e.g., MT1, MT2, and MT3) to broaden the immune response across multiple HIV-1 clades.
