Motexafin gadolinium + pemetrexed is a combination of two chemotherapeutic agents investigated primarily for the treatment of non-small cell lung cancer (NSCLC). Motexafin gadolinium (MGd) is a redox-active texaphyrin that disrupts redox-dependent pathways by inhibiting oxidative stress-related proteins, leading to apoptosis. It selectively targets tumor cells, interfering with energy metabolism and repair mechanisms, thereby sensitizing them to cell death. MGd also increases intracellular zinc levels in cancer cells, further disrupting enzyme activity and promoting apoptosis[1][6]. Pemetrexed is an antifolate antineoplastic agent that blocks folate-dependent enzymes involved in nucleotide synthesis, thereby inhibiting DNA and RNA synthesis essential for cancer cell proliferation[3][8]. The combination has been studied in clinical trials as second-line therapy for NSCLC after platinum-based regimens[1][2].
