Overview

Motexafin gadolinium + temozolomide is a combination therapy investigated primarily for the treatment of newly diagnosed supratentorial glioblastoma multiforme (GBM). Motexafin gadolinium is a synthetic metallotexaphyrin with radiosensitizing and chemosensitizing properties, acting as a redox-active agent that selectively targets tumor cells, inhibits cellular respiration, and may reduce radiation-induced oxidative damage repair. Temozolomide is an oral alkylating agent that induces DNA damage in tumor cells. The combination has been studied alongside standard fractionated radiotherapy to assess whether it improves overall survival and progression-free survival compared to historical controls. Clinical trials have shown the regimen to be well tolerated but without significant improvement in median overall survival over standard therapy[1][2][6].
