**Motixafortide + atezolizumab** is an investigational combination therapy that joins motixafortide, a cyclic peptide hematopoietic stem cell mobilizer and selective antagonist of C-X-C motif chemokine receptor 4 (CXCR4), with atezolizumab, a humanized IgG1 monoclonal antibody that targets programmed death-ligand 1 (PD-L1). Motixafortide facilitates mobilization of hematopoietic stem cells or augments anti-tumor immune responses by blocking CXCR4, thereby preventing retention of stem cells or immune cells within tissue compartments. Atezolizumab is an immune checkpoint inhibitor that blocks PD-L1 to restore T cell–mediated anti-tumor immunity. The rationale for combining these drugs is to enhance antitumor immunity: motixafortide may improve infiltration of immune cells into tumors, while atezolizumab removes inhibitory signals on T cells.[1][2][4][6]
