motolimod + nivolumab is a **combination drug regimen** composed of motolimod, a synthetic small molecule **toll-like receptor 8 (TLR8) agonist**, and nivolumab, a monoclonal antibody **immune checkpoint inhibitor targeting programmed death 1 (PD-1)**. The combination is investigated as an immunotherapeutic approach for squamous cell carcinoma of the head and neck (SCCHN). Motolimod stimulates innate and adaptive immune responses by activating TLR8, enhancing anti-tumor immunity, and promoting T helper cell type 1 polarizing cytokines. Nivolumab blocks PD-1, releasing the inhibition on T cells and restoring anti-tumor immune responses. Together, motolimod is hypothesized to augment the priming and activity of T cells and potentially synergize with the immune reactivating effects of PD-1 blockade by nivolumab. This regimen is not currently approved and remains investigational in phase 1b trials for SCCHN[2][4][6].
