mOTS-412 + anti PD-L1 is an investigational combination immunotherapy being developed by Bionoxx for the treatment of solid tumors. The regimen consists of mOTS-412, a proprietary oncolytic vaccinia virus, and a monoclonal antibody targeting Programmed death-ligand 1 (PD-L1). mOTS-412 is engineered to selectively infect and replicate within tumor cells, causing direct oncolysis and the release of tumor-associated antigens, which stimulates an innate immune response. The addition of a PD-L1 inhibitor is intended to block the interaction between PD-L1 on tumor cells and PD-1 on T cells, thereby overcoming immune suppression and enhancing the systemic, adaptive anti-tumor immune response initiated by the viral lysis. This dual approach aims to convert immunologically "cold" tumors into "hot" tumors, potentially improving therapeutic outcomes in patients with advanced malignancies. The program is currently in the preclinical stage of development.
