Overview

**mouse gp100 DNA vaccine** is a DNA-based cancer vaccine that encodes the murine (mouse) form of the melanoma-associated differentiation antigen gp100 (glycoprotein 100). As a DNA vaccine, it uses plasmid DNA encoding the gp100 antigen to induce an immune response, particularly cytotoxic T lymphocyte (CTL) responses, against cells expressing gp100, frequently melanoma cells. The vaccine is designed for immunotherapy of melanoma, leveraging xenogeneic (mouse) gp100 to break tolerance and enhance immunity against the human homolog. The vaccine may be delivered by various routes, commonly intramuscular injection, sometimes with electroporation, or (preclinically) orally via attenuated *Salmonella typhimurium.* Mouse gp100 DNA vaccine can be used alone or in combination with the human gp100 DNA vaccine in clinical trials to enhance immunogenicity. The primary mechanism is the induction of MHC class I-restricted, gp100-specific CD8+ T cell responses, resulting in tumor-protective immunity against melanoma[1][4]. \nMechanism of action: Induction of immune responses, specifically cytotoxic T cells, targeting the tumor antigen gp100; breaking self-tolerance to enhance anti-tumor immunity[1][4]. \nPrimary indication: Melanoma (including stage IIB–IV)[1][2]. \nDeveloper(s): Studies and trials have been conducted by research groups at Memorial Sloan Kettering Cancer Center and associated academic laboratories[1].
