Overview

The mouse tyrosinase-related protein 2 (mTRP-2) DNA vaccine is a xenogeneic cancer immunotherapy designed to treat malignant melanoma. It consists of a plasmid DNA vector encoding the murine version of the tyrosinase-related protein 2, an enzyme involved in melanin biosynthesis that is highly expressed in melanoma cells. The vaccine utilizes the principle of xenogeneic immunization; because the mouse TRP-2 protein is highly homologous but not identical to the human TRP-2 protein, it acts as a foreign antigen to circumvent the immune system's self-tolerance. Upon administration, the plasmid is taken up by host cells, leading to the expression of mTRP-2, which stimulates the production of cytotoxic T lymphocytes (CTLs) and antibodies that cross-react with the endogenous human TRP-2 on tumor cells, ultimately promoting tumor regression.
